This is a small part of a larger project which is to understand the role of Epstein Barr virus (EBV) in lymphoma-genesis. The studies are performed on two types of cell-lines, CBMI-Ral-STO is “immortal” but does not show fully malignant phenotype and RAEL cell-line can form colonies in soft agarose and generate tumors. One of the key roles will be studying transcription factors that may regulate the OCT-2, which is a product of POU2F2 gene. It is believed that OCT-2 with EBNA-1 may mutually regulate Cp and Qp which are the promoters of EBV, and expression of OCT-2 in CBMI-Ral-STO is much lower than in RAEL cells. Most of studies emphasized on studying what OCT-2 regulates, but there are little known about what regulates OCT-2. Also we will look to other proteins that may regulate expression of two cells and finally we go through the ontology of two cell lines and look to the difference between the two types. In the process of the work we have selected 5 transcription factors EGR2, Smad3, FOXJ2, TCF-7, and RXR alpha. These proteins may regulate expression of OCT-2 in the cell. Thus we have studied pathways in which they are involved also we have brought other proteins with which these 5 transcription factors interact. Also this work shows the difference in molecular functioning, biological process and cellular compartment between two cell lines.
